3. Product description:

PI is a nuclear staining reagent that can stain DNA. It emits red fluorescence after intercalating double-stranded DNA. It is commonly used for the detection of apoptosis or necrosis, and it is often used for analysis by flow cytometry. Although PI cannot pass through the living cell membrane, it can pass through the damaged cell membrane to stain the nucleus. PI is often used in combination with Calcein, AM, Hoechst 33258 or Hoechst 33342, etc. It can stain live and dead cells simultaneously. The excitation and emission wavelengths of the PI-DNA complex are 535nm and 615nm, respectively.

4. Dyeing process

(1) Prepare 10-50 μM PI solution with PBS or appropriate buffer.

(2) Add 1/10 of the culture medium volume of PI solution to the cell culture medium (1/10 concentration of PI buffer solution can also be used instead of the culture medium).

(3) Incubate the cells at 37 ° C for 10-20 minutes.

(4) Wash the cells twice with PBS or a suitable buffer.

(5) Observe the cells using a fluorescence microscope with an excitation wavelength of 535nm and an emission wavelength of 615nm.
